The Hampshire golfer stood three shots back at the start of play, trailing overnight leader Bubba Watson.
But, with Watson fading, world number one Rory McIlroy's late charge falling short and Tiger Woods retiring with an Achilles injury , Rose held his nerve.
A final round two-under 70 was enough to win by a shot from Watson, who missed a 10-foot birdie putt at 18.
That followed a fantastic recovery from a wayward drive down the last by Watson, from almost the same spot where Rose had bogeyed to open the door for the American 15 minutes earlier.
"There's been a lot of hard work going on in my game," said Rose. "And days like this make it all worthwhile.
"I really enjoyed playing with Bubba Watson for three days. And going out one group ahead of the leaders is never a bad thing.
"But the two shots he hit into 18 had me sweating."

'Locked-in syndrome' man to have right-to-die case heard

Tony Nicklinson, who is paralysed and wants a doctor to be able to lawfully end his life, should be allowed to proceed with his "right-to-die case", a High Court judge has ruled.
The 58-year-old from Melksham, Wiltshire, has "locked-in syndrome" following a stroke in 2005 and is unable to carry out his own suicide.
His is seeking legal protection for any doctor who helps him end his life.
The Ministry of Justice argues making such a ruling would change murder laws.
"Locked-in syndrome" leaves people with paralysed bodies but fully-functioning minds.
Mr Nicklinson, who communicates through the use of an electronic board or special computer, says his life is "dull, miserable, demeaning, undignified and intolerable".
His wife Jane has said that "he just wants to know that, when the time comes, he has a way out".
"If you knew the kind of person that he was before, life like this is unbearable for him," she added.
She said she did not know when her husband might actually want to die. "I suppose just when he can't take it any more," she said.

The death has been announced of Sherwood Rowland - the US chemistry professor who first suspected that the Earth's protective ozone layer was being depleted by man-made chemicals.
Prof Rowland published a paper on the dangers of the then-popular chlorofluorocarbons - CFCs - in 1974.
His ideas were derided at the time by other scientists and especially the chemical industry.
But 20 years later, Sherwood Rowland won a Nobel Prize for his work.
