Trent Hodkinson and James Graham will co-captain the side. Ennis had held on slight hope of a miracle recover after breaking two bones in his foot in Canterbury's 18-12 preliminary finals win against the Panthers.
It will be the second time injury has cruelled Ennis' premiership chances having missed Brisbane's 2006 grand final after rupturing his anterior cruciate ligament earlier that season. Canterbury coach Des Hasler it yet to name a replacement for Ennis with Moses Mbye, Reni Maitua, Josh Jackson and Josh Reynolds dummy half options.
